Simpson clocked a wind-assisted 11.04 seconds to win the women's sprint and Mullings clocked a decent 10.07 seconds into a headwind to land the men's 100 after posting a meet record 10.05 in the heats against a -1.2 metres per second wind.
On the comeback trail after injury, Simpson followed up her win at the Padua International on Sunday by snatching a narrow win over Trinidad & Tobago's Kelly-Ann Baptiste (11.10), pushed by a 2.8 metres per second wind.
"I did not expect this fast race today," said Simpson, who is slated to run the 100m at the Brussels Golden League today.
Britain's Emma Ania (11.21) and Jamaica's Berlin IAAF World Championship relay gold medallist, Aleen Bailey (11.23) were third and fourth, resepctively.
Mullings, who also celebrated sprint relay gold in Berlin with Jamaica's men's squad, led a Caribbean capture of all the podium spots in the men's 100 metres.
Against a wind of minus 1.5 mps, he was a clear winner over fellow Jamaican Lerone Clarke (10.13), with Antigua and Barbuda's Brendan Christian third in 10.27.
Barbados champion Andrew Hinds was seventh in 10.42 seconds.
There was a top-three finish for Jamaica in the 100-metre hurdles with Nickeisha Wilson (13.03) taking the runner-up spot behind American winner Damu Cherry (12.97).
Bahamian Olympic triple jump bronze medallist, Leevan Sands, made an appearance in the long jump and placed sixth at 7.58 metres, just behind fifth-placed Bermudian Tyrone Smith (7.66m) as Ukraine's Andriy Makarchev topped the field at 7.90 metres.
Russian Olga Kucherenko won the women's long jump at 6.76 metres, with Jamaican Jovanee Jarrett seventh with 6.40m.
