Australia’s left-arm pacer Mitchell Starc bowled debutant Kevlon Anderson, who was also asked to stand in as opener, for three.
Earlier, Steven Smith (48) and Cameron Green (46) led the way for Australia after they won the toss and opted to bat first in conditions which favoured the quicker bowlers.
West Indies pacers Shamar Joseph (4-33), Jayden Seales (3-59) and Justin Greaves (3-56) made good use of the significant sideways movement — which was amplified under the floodlights — off the surface.
The West Indies make-shift openers predictably found the going tough for the nine overs remaining in the evening against the high-quality bowling attack.
West Indies trail 0-2 in the three-Test series after losing the previous matches in Barbados and Grenada.
